I've been playing Gran Turismo since GT5 for couple of year and I really enjoy the game. So when GT6 came out I have to have it and it surprise me that Nitro is one of the tuning option !!!
I tried it on my fully tuned Nissan GT-R '12 with Racing Hard tire. I did one lap in Tsukuba without Nitro and clocked 52'804. For the third lap I used Nitro in all the corner exit, straight ... and the result is 52'105 not as dramatic as I thought !!!. That time difference could be just mean I took a better line than the other lap.
Could someone explain to me how Nitro work in GT6 ???

I haven't used nitrous yet, but the key to making the most out of it is in the RPM range you use it in. It's best to use nitrous in a low gear so you can benefit from a greater power gain. On the track, it would be best to use Nitrous after exiting a tight corner (as long as you've got traction).

You can set in the tuning options to allow more power to be used. Try cranking that and using it in the correct RPM range.

You would also benefit from the nitrous a lot more on a track that isn't so short that has a high brake to full throttle ratio.

Silverstone GP circuit for example has 5 decent length straights, nitrous here would make a huge difference (for a lap or two anyway).
